QUICK AND EASY” genomic prep for PCR analysis
From Steve Pollock, Louisiana State University, March 3, 2003
To quickly and easily screen one or several strains of Chlamydomonas the following protocol works amazingly well:
1) resuspend a pellet of approximately 5-10 uL in size in 50 uL of 10mM NaEDTA by vortexing.
2) incubate at 100C for 5 minutes (thermocycler works well).
3) vortex to mix.
4) centifuge at 12000g for 1 minute.
5) use 1.5 uL in a 25 uL PCR reaction.
